Salvatore Storti, his maternal great-great-grandfather, arrived in America from Italy in 1904 and finally settled in Pennsylvania. In 1917, Luigia Colucci followed her husband to the United States.

She was his great-great-grandmother. DeSantis' father installed Nielsen TV rating boxes, while his mother worked as a nurse. When he was six years old, his family relocated from Orlando, Florida, to Dunedin, Florida.

Christina Marie DeSantis, his sister, was born on May 5, 1985, in Orlando and passed away in 2015. He played on the Dunedin National Little League squad that advanced to the Williamsport, Pennsylvania, Little League World Series in 1991.

DeSantis graduated in 1997 from Dunedin High School after attending Our Lady of Lourdes Catholic School.
